Overview

This application note demonstrates how circular dichroism spectroscopy and differential scanning calorimetry can be used together to investigate the structure and thermal stability of lipid nanoparticles for mRNA delivery. These complementary, label-free techniques provide insight into RNA secondary structure, lipid phase behaviour, and interactions within mRNA lipid nanoparticle assemblies. The combined approach supports formulation development, comparability assessment, and improved understanding of factors that influence stability and performance of mRNA lipid nanoparticle drug products.
